What it means
Unlike semaglutide and tirzepatide, orforglipron is not built from amino acids. It is a small-molecule drug that still switches on the GLP-1 receptor. That design lets it be absorbed from a normal tablet, taken any time of day, with or without food.
The FDA approved it as Foundayo on April 1, 2026, for chronic weight management in adults with obesity, or overweight with at least one weight-related condition, alongside diet and activity. In the ATTAIN-1 trial, average weight loss at 72 weeks was about 12.4% on the highest dose, compared with about 0.9% on placebo.
The label starts at a low dose and steps up at least every 30 days. Its half-life is roughly 29 to 49 hours, which supports once-daily dosing. It carries the same boxed warning about thyroid C-cell tumors as other GLP-1 medicines.
Stomach side effects such as nausea, constipation and diarrhea were the most common in trials.
Why it matters
Orforglipron is the first GLP-1 pill for weight without the empty-stomach routine that oral semaglutide needs.
